Specifications for a Light Sheet Fluorescence Microscope (LSFM) for the application with live and optically cleared biological samples.
1. Product Description: Light Sheet Fluorescence Microscope (LSFM) system capable to image live and optically cleared sample, fully automated and computer controlled. The instrument will be part of a multidimensional imaging facility in RCSI.
2. Product Requirement: This product is required for live sample fluorescence imaging and imaging of fixed specimen, which will be mostly optically cleared. Particular applications will use live and fixed samples expressing one or more GFP fusion proteins, which in the case of live samples may act as fluorescence resonance energy transfer partners. The equipment should allow long term observation of living samples, which requires the control of incubation parameters inside the imaging chamber (temperature, CO₂, O₂, perfusion). The application has to be flexible with regards to the spectra of the probes used. Therefore a big number of different beam splitters and filters and the possibility of an on-site exchange or other technical solutions to increase flexibility and sensitivity are appreciated.
